2-MeTHF | Bio-Based Aprotic Solvent for Pharmaceutical Synthesis

Product HighlightPharmaceuticals

A high-purity (≥99.9%) alternative to tetrahydrofuran (THF), produced from agricultural waste residue and available in ISO tank, drum and IBC pack sizes for Grignard, cross-coupling and peptide synthesis chemistries.

Key benefits of 2-MeTHF include:

  • Improved chemical stability: Increased stability to acid and base compared with THF, supporting safer handling across a broader range of reaction conditions.
  • Reverse water solubility: Solubility in water falls from 14 wt% at 20°C to 6.6 wt% at 60°C, allowing more effective phase separation at elevated temperature and simplifying aqueous work-up.
  • Efficient solvent recovery: A favourable solvent-free azeotrope (10.6% water) means a single fractionation step is sufficient for complete recovery and drying, in contrast to the more complex recovery THF requires.
  • Safe classification: no classification as a carcinogen, mutagen, or reproductive toxicant.
  • ICH Q3C Classification: Class 3 (solvents with low toxic potential, permitting higher allowable residual levels in pharmaceutical drug substances and APIs).

Why manufacturers are re-evaluating THF-based solvent systems

Grignard reactions, lithiations and a range of cross-coupling chemistries in Active Pharmaceutical Ingredient (API) and intermediate synthesis have long relied on tetrahydrofuran (THF) as the standard aprotic solvent. THF's chemical instability toward acids and bases, and a water azeotrope that complicates recovery and drying all add operational and safety burden to processes built around it.

As process chemists look for aprotic solvents that retain THF's solvency while improving on handling, recovery economics and feedstock profile, interest has grown in alternatives produced from agricultural by-products rather than petrochemical feedstocks.

2-MeTHF: A successful partnership between IMCD and QQChem

Over 20 years ago, Jiangsu Qingquan Chemical, better known as QQChem, set out to challenge petrochemical norms by developing a renewable solvent from agricultural waste. The result was 2-methyltetrahydrofuran (2MeTHF), derived from furfural, long before ‘green chemistry’ became an industry buzzword.

In 2005, IMCD recognised the potential and partnered with QQChem. What began as a shared commitment to innovation has grown into a trusted, global partnership that continues to redefine how the pharmaceutical industry approaches sustainability.

In 2018, the IMCD Synthesis team launched the first Greener Solvents Seminar, with 2-MeTHF emerging as one of the most promising alternatives. Since then, IMCD has reached more than 1500+ chemists worldwide, supporting our customers with their solvent selection.

Today, 2-MeTHF is a well-established solvent among chemists. However, there are still valuable technical insights, practical tips, and best practices that deserve greater attention. That’s why we’ve created a new IN FOCUS Session: Unlocking the Potential of 2-MeTHF. This seminar explores a range of synthetic transformations in which (2-MeTHF) demonstrates clear advantages over conventional THF and show how 2-MeTHF is not only a renewable, biomass-derived alternative to THF, but also a low-toxicity, safer solvent option that can significantly reduce environmental impact.

Applications of 2-MeTHF across pharmaceutical synthesis

The 2-MeTHF range brings benefits to the following pharmaceutical synthesis applications:

  1. Organometallic chemistry: As a Grignard reaction and lithiation solvent, 2-MeTHF's improved chemical stability and reverse water solubility support more straightforward handling and phase separation than THF-based systems.
  2. Peptide synthesis: Used as an extraction solvent in solution- and solid-phase peptide synthesis.
  3. Bi-phasic reactions: Its inverse water solubility at elevated temperature supports use as a phase-transfer medium in bi-phasic reactions, alongside application in cinchona alkaloid-mediated chiral induction and enzymatic chemistry.
  4. Reduction chemistry: Applied in chelation-controlled reductions and nickel sponge reduction reactions
  5. Cross-coupling reactions: Suitable for Suzuki, Heck, Stille, Sonogashira, Negishi, Kumada and Buchwald couplings, including iron-catalysed cross-coupling routes, for carbon-carbon and carbon-nitrogen bond formation.
  6. N-alkylation and Wittig reactions: Supports N-alkylation of isatin analogues and double-bond formation in Wittig reactions.
  7. Chemical properties: Increase chemical stability to acid & base versus THF improved safety.

2-MeTHF specification, packaging and supply through IMCD

  • Controlled purity and peroxide risk: Supplied at a minimum 99.9% purity with a maximum water content of 0.03% (300 ppm) and BHT antioxidant control limiting peroxide content to a maximum of 3 ppm.
  • Additional grades: Tocopherol-stabilized (Vitamin E) grade, requested by the flavours & fragrance industry, and an ultra-low-water anhydrous grade, for moisture sensitive chemistries.
  • Bio-based manufacturing route: Produced from agricultural waste residue rather than petrochemical feedstock, supported by two dedicated manufacturing plants in China with a combined annual capacity of 30 kilotonnes (KT).

2-MeTHF is available in the following pack sizes:

  • ISO tank: 18–25 MT
  • Drums: 160 kg net, 200L
  • IBC: Available on request

IMCD and QQChem have built a robust supply network in Europe, India, and the US offering packaging from 1L samples to bulk isotanks. Sustainability performance is independently assessed on both sides of the partnership and both companies have been awarded with EcoVadis medals for their sustainability performance.
